Highly dynamic reorganization of plasma membrane and cytoskeleton can be modulated by emerging lipid rafts-modulating proteins in response to external cues. Structural analysis of Annexin II, a calcium-dependent anionic phospholipid binding protein, revealed that membrane binding surface of the annexin had several cationic patches for binding to PtdIns(4,5)P2, by which the annexin may induce clustering of PtdIns(4,5)P2 in a calcium-dependent manner, remodeling of lipid rafts, and thereby, reorganization of membrane-cytoskeleton. Localized reorganization of lipid rafts may break a symmetry in cell membrane and may play roles in polarized cellular processes, including development, directional migration, and cell-cell adhesion. Time-lapse cell imaging and SPR(surface plasmon resonance) measurement demonstrated that annexin II binds specifically to PtdIns(4,5)P2. Imaging study using partial dominant negative mutant annexins unable to bind to PtdIns(4,5)P2 showed that annexin II may play a crucial role in regulating cell polarity in SDF1-induced chemotactic T cells and cell-cell adhesion in epithelial cells.
